Characterization of Borrelia Burgdorferi Gene Products Critical for Pathogen Persistence and Transmission

چکیده
B. burgdorferi, the pathogen of Lyme borreliosis, persists in a natural tick-rodent transmission cycle. Assessment of in vivo and in vitro microbial transcriptome encoding selected membrane proteins by high throughput qRT-PCR, showed bba62, bba52, bb0323, bba74, bbe31, bbg01 are predominantly expressed during vector-specific phase of B. burgdorferi life cycle compared to mammalian host. In addition, bba52 and bba62 are expressed exclusively in vector, and have no detectable expression in the mammalian host. Further assessment of these proteins may shed new light on vector-pathogen interaction and B. burgdorferi persistence through a tick-rodent cycle.

Borrelia burgdorferi small lipoprotein Lp6.6 is a member of multiple protein complexes in the outer membrane and facilitates pathogen transmission from ticks to mice
Borrelia burgdorferi lipoprotein Lp6.6 is a differentially produced spirochete antigen. An assessment of lp6.6 expression covering representative stages of the infectious cycle of spirochetes demonstrates that the gene is solely expressed during pathogen persistence in ticks. Deletion of lp6.6 in infectious B. burgdorferi did not influence in vitro growth, or its ability to persist and induce i...

Borrelia burgdorferi, the pathogen of Lyme borreliosis, persists in nature through a tick-rodent transmission cycle. A selective assessment of the microbial transcriptome, limited to gene-encoding putative membrane proteins, reveals that bba52 transcription in vivo is strictly confined to the vector-specific portion of the microbial life cycle, with the highest levels of expression noted in fee...
متن کاملExpression and Purification of Recombinant Outer Surface Protein D of Borrelia burgdorferi
To carry out the immunological experiments on the serum of Multiple Sclerosis (MS) patients, based on a correlation between Borrelia burgdorferi infection and contracting MS autoimmune disease the outer surface protein D (OspD) of the bacterium was expressed and purified. A clone containing the OspD gene in pET11a expression vector under the control of T7 promoter was transformed to the bacteri...
متن کاملBB0323 and novel virulence determinant BB0238: Borrelia burgdorferi proteins that interact with and stabilize each other and are critical for infectivity.
We have shown that Borrelia burgdorferi gene product BB0323 is essential for cell fission and pathogen persistence in vivo. Here we describe characterization of a conserved hypothetical protein annotated as BB0238, which specifically interacts with the N-terminal region of BB0323. We show that BB0238 is a subsurface protein, and similar to BB0323, exists in the periplasm and as a membrane-bound...

Borrelia burgdorferi, the spirochetal agent of Lyme borreliosis, is predominantly transmitted by Ixodes ticks. Spirochetes have developed many strategies to adapt to the different environments that are present in the arthropod vector and the vertebrate host. This review focuses on B. burgdorferi genes that are preferentially expressed in the tick and the vertebrate host, and describes how selec...
